I am trying to:
I want manage user role access. When I edit capability settings to an existing role or a custom role I created, users under the affected role can't log into site admin and get HTTP ERROR 500. The particular settings are related to Pages access.
I was using another user role access plugin in conjunction with Toolset Access plug. After realizing the problem, I turn off the other potential conflicting plugin and still get access error. After elimination process, I find that by disabling Toolset Access plugin, the problem goes away.

The problem you mentioned above is abnormal, please check these:
1) Make sure you are using the latest version of Toolset Access plugin, you can download it here:

2) In case it is a compatibility problem, please deactivate other plugins, and switch to wordpress default theme 2019, deactivate all custom PHP code, and test again
3) Also check if there is any PHP/JS error in your website:

Thanks for the quick response.
It sounds like this will take a good few hours to narrow down the cause as the possibility can be in many places. I'm under a tight deadline. To immediately resolve the issue, I'm using another plugin, which works fine, for similar functions and have to disable Toolset Access plugin.

500 error means there are some PHP errors in your website, it is helpful to debug this problem.
